組蛋白 (Histone H3) 介紹
染色質(zhì)結(jié)構(gòu)的調(diào)節(jié)在真核生物的轉(zhuǎn)錄調(diào)控中起著重要作用。核小體是染色質(zhì)的主要組成部分,由DNA 纏繞在組蛋白周圍形成。核小體是一個組蛋白八聚體,由 147 個堿基對的 DNA 和 8 個核心組蛋白組成,包含 H2A、H2B、H3 和 H4 分子各兩個,組成一個 H3-H4 異四聚體和兩個 H2A-H2B 異二聚體。核小體像珠串一樣串連在一起,并被包裝成更高階的染色質(zhì)結(jié)構(gòu)。核心組蛋白的n端末端經(jīng)歷不同的翻譯后修飾,包括乙?;?、磷酸化和甲基化,這些修飾是對細(xì)胞信號刺激的反應(yīng),對基因表達(dá)有直接影響。在大多數(shù)物種中,組蛋白 H2B 主要在賴氨酸 5、12、15 和 20 位點(diǎn)發(fā)生乙酰化。組蛋白 H3 主要在賴氨酸 9、14、18 和 23 位點(diǎn)發(fā)生乙?;Y嚢彼?9 位點(diǎn)的乙?;谀承┥锏慕M蛋白沉積和染色質(zhì)組裝中具有主導(dǎo)作用。在有絲分裂和減數(shù)分裂中,組蛋白 H3 的 Ser10 位點(diǎn)的磷酸化與染色體凝聚緊密相關(guān)。
核小體將 DNA 包裹并壓縮到染色質(zhì)中,這限制了 DNA 對需要 DNA 作為模板的細(xì)胞機(jī)制的可及性。因此,組蛋白在轉(zhuǎn)錄調(diào)節(jié)、DNA 修復(fù)、DNA 復(fù)制和染色體穩(wěn)定中發(fā)揮著核心作用。DNA 可及性是通過組蛋白翻譯后修飾(也稱為組蛋白編碼)和核小體重構(gòu)的復(fù)雜組合來調(diào)控的。H3 完全通過 DNA 復(fù)制偶聯(lián)途徑沉積到染色質(zhì)中,該途徑可與減數(shù)分裂同源重組過程中的 DNA 復(fù)制或 DNA 修復(fù)合成相關(guān)。
