產(chǎn)品介紹 The protein encoded by this gene has been termed a 'growth' or 'plasticity' protein because it is expressed at high levels in neuronal growth cones during development and axonal regeneration. This protein is considered a crucial component of an effective regenerative response in the nervous system. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]

Function:
This protein is associated with nerve growth. It is a major component of the motile 'growth cones' that form the tips of elongating axons. Plays a role in axonal and dendritic filopodia induction.

Subunit:
Identified in a complex containing FGFR4, NCAM1, CDH2, PLCG1, FRS2, SRC, SHC1, GAP43 and CTTN. Binds calmodulin with a greater affinity in the absence of Ca(2+) than in its presence.

Cell membrane; Peripheral membrane protein; Cytoplasmic side. Cell projection, growth cone membrane; Peripheral membrane protein; Cytoplasmic side. Cell junction, synapse. GAP43 (Growth associated protein-43)又稱作neuromodulin,是一個軸突膜蛋白,是一種神經(jīng)特異性的蛋白質(zhì),參與神經(jīng)細胞外生長及突觸發(fā)育形成和神經(jīng)細胞再生。在神經(jīng)元發(fā)育和再生過程中以高水平表達。能調(diào)解軸突延伸作用,改變細胞形態(tài)。作為細胞內(nèi)信號,可大大增強與G蛋白偶聯(lián)的受體轉運作用。
神經(jīng)生長相關蛋白-GAP-43和神經(jīng)細胞黏附因子(neural cell adhesion molecule,NCAM)與突觸可塑性密切相關.GAP-43是一種神經(jīng)細胞膜上的特異性磷蛋白,在神經(jīng)發(fā)育和再生過程中呈現(xiàn)高表達,被作為突觸生長的標志物,有稱脊髓生長相關蛋白
GAP-43與CaM結合,參與G蛋白相互作用,神經(jīng)遞質(zhì)的釋放,作用于胞吞/胞吐過程,通過小囊溶合或誘導生長錐和突觸前末端的胞吞促進膜擴展,與海馬長時程增強密切相關.
神經(jīng)細胞黏附因子是細胞表面糖蛋白大家族的成員之一,促進軸突生長,對長時記憶的保持有重要影響,同時,GAP-43對其具調(diào)節(jié)作用,廣泛而深入地研究GAP-43意義深遠。
