Database links: GenBank: AAP93872.1 Important Note: This product as supplied is intended for research use only, not for use in human, therapeutic or diagnostic applications. 天蠶素(cecropins)是第一個(gè)被發(fā)現(xiàn)的動(dòng)物抗菌肽,1980年,由Boman等從天蠶蛹中分離得到。該類多肽抗生素一般含有37~39個(gè)氨基酸殘基,不含半胱氨酸,其N端區(qū)域具有強(qiáng)堿性,可形成近乎完美的雙親螺旋結(jié)構(gòu),而在C端區(qū)域可形成疏水螺旋,兩者之間有甘氨酸和脯氨酸形成的鉸鏈區(qū),多數(shù)多肽的C端被酰胺化,酰胺化對(duì)其抗菌活性具有重要作用。天蠶素(cecropins)對(duì)革蘭氏陽(yáng)性菌、部分革蘭氏陰性菌具有很強(qiáng)的殺傷力,而對(duì)真菌和真核細(xì)胞沒(méi)有毒性。目前研究認(rèn)為,抗菌肽是最具潛力的抗生素替代品之一。而天蠶素又是目前研究比較清楚的抗菌肽。 天蠶素抗菌肽利用N端所帶的正電荷與原核細(xì)胞的細(xì)胞膜磷脂雙分子層密集的負(fù)電荷靜電相吸,C端柔性的插入疏水區(qū),在兩親性α-螺旋作用下,在原核細(xì)胞膜上形成4nm的離子通道,離子通道的形成改變了細(xì)胞膜內(nèi)外的滲透壓,細(xì)胞膜內(nèi)物質(zhì)大量外滲,致細(xì)菌細(xì)胞死亡,起到殺菌作用。天蠶素抗菌肽對(duì)細(xì)菌、病毒、念珠菌、原蟲(chóng)、癌細(xì)胞具有廣譜的活性,能抑制革蘭氏陽(yáng)性和陰性球菌和桿菌,如綠膿桿菌、大腸桿菌、傷寒桿菌、痢疾桿菌、布氏桿菌、金黃色葡萄球菌、溶血性鏈球菌等等,并對(duì)感染植物的多種病原菌也有明顯的殺菌作用。另外,天蠶素抗菌肽對(duì)高度耐藥的傷寒桿菌、耐藥性金葡菌同樣敏感,對(duì)感冒、疤疹病毒能抑制其繁殖,對(duì)瘧原蟲(chóng)、痢疾阿米巴均有效 |
